AUTHENTICATION OF COMMERCIAL CHILEAN MOLLUSKS USING RIBOSOMAL INTERNAL TRANSCRIBED SPACER (ITS) AS SPECIE-SPECIFIC DNA MARKER Gayana
Aguilera-Muñoz,Felipe; Valenzuela-Muñoz,Valentina; Gallardo-Escárate,Cristian.
Contemporary world food safety standards have emphasized the implementation of efficient food traceability systems, including the correct labeling of products for the authentication of the commercialized species. In this context, Chile has reached an important development in agricultural exportations, reaching near US$ 3.82 billion in 2007. Since the identification based on morphologic and organoleptic characteristics is complex during the elaboration process of aquaculture products, molecular tools need to be developed in order to genetically trace aquaculture products. This study analyzed five species of Chilean commercial mo Husks using molecular DNA markers. From the partial sequences of the ribosomal region ITSl-5.8SrDNA-ITS2, the specific primers for...

ESTUDIOS MORFO-ANATÓMICOS, CITOGENÉTICOS Y MOLECULARES EN Talinum (TALINACEAE): UNA NUEVA ENTIDAD PARA EL NOROESTE ARGENTINO Acta biol.Colomb.

Genetic diversity, paraphyly and incomplete lineage sorting of mtDNA, ITS2 and microsatellite flanking region in closely related Heliopora species (Octocorallia) ArchiMer

Examining genetic diversity and lineage sorting of different genes in closely related species provide useful information for phylogenetic analyses and ultimately for understanding the origins of biodiversity. In this study, we examined inter- and intraspecific genetic variation in internal transcribed spacer 2 (ITS2), partial mitochondrial gene (mtMutS), and nuclear microsatellite flanking region in two closely related octocoral species (Heliopora coerulea, HC-A and HC-B). These species were recently identified in a population genetic study using microsatellite markers. The two species have different reproductive timing, which ecologically promotes lineage sorting. In this study, we examined whether species boundaries could be detected by the commonly used...

Intraespecific variation on the aedeagus of Anopheles oswaldoi (Peryassú) (Diptera: Culicidae) Neotropical Entomology
Motoki,Maysa T; Santos,Cecília L S dos; Sallum,Maria A M.
Anopheles (Nyssorhynchus) oswaldoi (Peryassú) comprises a species complex in South America. Specimens from two localities in east Mata Atlântica were characterized both at the morphological and molecular level as An. oswaldoi s.s. Intraspecific variation of the shape of the apex of the aedeagus of the male genitalia of specimens of Anopheles (Nyssorhynchus) oswaldoi s.s. from Vale do Ribeira, Mata Atlântica, São Paulo State, Brazil, was observed. Distinctive aedeagus of the specimens from Vale do Ribeira, Mata Atlântica, were evaluated, illustrated and compared to that of An. oswaldoi s.s.

PCR-RAPD and PCR-RFLP polymorphisms detected in Anopheles cruzii (Diptera, Culicidae) Rev. Bras. entomol.
Calado,Daniéla; Navarro-Silva,Mario Antonio; Sallum,Maria Anice Mureb.
The aim of the present study was to examine genetic variability in populations of An. cruzii by employing PCR-RAPD and PCR-RFLP markers. All analyses were carried out using individuals of the F1 generation of wild caught females obtained in Santa Catarina State (Florianópolis and São Francisco do Sul), Paraná State (Morretes, Paranaguá and Guaratuba) and São Paulo State (Cananéia). In the PCR-RAPD experiments, seven primers were used for comparisons within and among populations. The restriction profile of the ITS2 including a fragment of both 5.8S and 28S regions of the rDNA was obtained with the enzymes BstUI, HaeIII, TaqI, HhaI, Sau96I, HinfI, HincII and NruI. The PCR-RAPD method detected a large number of polymorphic bands. Genetic distance among...

Secondary structure of nrDNA Internal Transcribed Spacers as a useful tool to align highly divergent species in phylogenetic studies Genet. Mol. Biol.
Giudicelli,Giovanna C.; Mäder,Geraldo; Silva-Arias,Gustavo A.; Zamberlan,Priscilla M.; Bonatto,Sandro L.; Freitas,Loreta B..
Abstract Recently, it has been suggested that internal transcribed spacer (ITS) sequences are under selective constraints to preserve their secondary structure. Here, we investigate the patterns of the ITS nucleotide and secondary structure conservation across the Passiflora L. genus to evaluate the potential use of secondary structure data as a helpful tool for the alignment in taxonomically complex genera. Considering the frequent use of ITS, this study also presents a perspective on future analyses in other plant groups. The ITS1 and ITS2 sequences presented significant differences for mean values of the lowest energy state (LES) and for number of hairpins in different Passiflora subgenera. Statistical analyses for the subgenera separately support...
